    天门冬叶状枝原生质体的分离制备

    Isolation of protoplasts from cladodes of Asparagus cochinchinensis

    • 摘要: 本研究以天门冬叶状枝为材料,分析了酶浓度组合和酶解时间对天门冬原生质体制备的影响,进一步采用双因素正交试验优化制备体系.结果表明,酶浓度组合为1.5%纤维素酶+0.8%离析酶,酶解3 h为最佳条件,可获得最高的原生质体总产量及有活性原生质体数量,这为天门冬原生质体高效制备提供了重要的理论依据.

       

      Abstract: This study utilized cladodes of Asparagus cochinchinensis to investigate the effects of enzyme concentration and hydrolysis time on protoplast isolation. A dual-factor orthogonal experiment was subsequently conducted to optimize the isolation protocol. The results demonstrated that optimal conditions, consisting of 1.5% Cellulase, 0.8% Macerozyme and 3 h of hydrolysis, yielded the highest values for both total protoplast count and viable protoplast numbers. This provides important theoretical support for efficient protoplast isolation from A. cochinchinensis.
