    Synergistic Extraction of Totalflavonoids from Pomegranate Peel in Panzhihua by Joint Use of Cellulase and Ultrasonic Wave

    • 摘要: 研究纤维素酶联合超声波协同提取四川攀枝花石榴皮总黄酮.探究了乙醇体积分数、料液比、酶活量、超声波功率、提取时间、提取温度等对总黄酮产率的影响.以正交试验设计优化出了最佳提取条件:酶活度为20U/mL、超声波功率为280 W、提取时间为20min、提取温度为60℃, 此条件下总黄酮产率为8.805%, 提取物对1, 1-二苯基-2-三硝基苯肼自由基的清除能力强于维生素C和芦丁, 其清除能力存在较好的量效关系.

       

      Abstract: To study the synergistic extraction of flavonoids in Panzhihua pomegranate peel by ultrasonic-assisted cellulase method, the effects of ethanol volume fraction, solid-liquid ratio, enzymatic activity, ultrasonic power, extraction time and extraction temperature on the yield rate of total flavonoids are examined.By orthogonal experiment design, the optimal extraction conditions were acquired:when the enzyme activity is 20 U/mL, ultrasonic power is at 280 W, extraction time lasts 20 minutes, extraction temperature at 60℃, the yield rate of total flavonoids could reach 8.805%.The scavenging ability of extract for 1, 1-diphenyl-2-picrylhydrazyl free radical was stronger than vitamin C and rutin and the scavenging ability was of good dose-effect relationship.The experiment method can serve as a good means for the better utilization of Panzhihua pomegranate peel resource.
