    Redescription and Molecular Phylogeny of Zschokkella parasiluri Fujita, 1927

    • 摘要: 为确认鲇楚克拉虫Zschokkella parasiluri Fujita, 1927的物种有效性, 对采自重庆市嘉陵江段的该物种进行了形态学重描述和分子系统学的研究.基于形态特征和18SrDNA对鲇楚克拉虫及其相似种进行变异位点、遗传距离、序列相似度和系统发育分析.鲇楚克拉虫孢子壳面观呈卵圆形, 壳瓣上有711条与缝嵴平行的条纹, 缝嵴呈“S”形.孢子长11.84±0.32 (11.3312.60) μm, 孢子宽5.44±0.15 (5.095.78) μm.两个极囊等大, 呈球形, 分布于孢子两端, 极囊直径3.82±0.24 (3.454.22) μm, 两极囊间距2.60±0.23 (2.232.87) μm, 极丝明显, 盘曲56圈.所获鲇楚克拉虫 (登陆号为:MH517584) 与GenBank库中的鲇楚克拉虫湖北种群 (登陆号为:DQ377689) 的序列相似度为99%, 遗传距离为0.004, 两种群序列仅有8个变异位点.重庆为该物种的新分布地区;两极虫科粘孢子虫由海水支系和淡水支系构成, 两极虫属和楚克拉虫属混合聚支而表现出两者的非单系性和较近的亲缘关系.

       

      Abstract: In order to confirm the validation of Zschokkella parasiluri Fujita, 1927, redescription and molecular phylogeny on Z.parasiluri collected from Jialing River, Chongqing, China.were conducted, Comparative analyses on Z.parasiluri and related species were conducted based on morphological characteristics, 18 SrDNA variation sites, genetic distances, sequence similarities, phylogenetic analysis.The results indicated that the spore shape of Z.parasiluri was oval with 711 striations paralleling to the suture ridge (“S”form) on the surface of shell valves.Its spore was 11.84±0.32 (11.3312.60) μm long, and 5.44±0.15 (5.095.78) μm wide, each spore had two equal spherical polar capsules 3.82±0.24 (3.454.22) μm in diameter, and positioned at both ends of the spore.The distance between the two polar capsule was 2.60±0.23 (2.232.87) μm and the polar filaments were obvious and coiled 56 turns.The results also indicated that the sequence similarity between the Chongqing population of Z.parasiluri (GenBank accession no.MH517584) and the Hubei population of Z.parasiluri (GenBank accession no.DQ377689) was 99%, and the genetic distance of them was 0.004.There were only 8 variable sites between the sequences of the two populations.There was no record of Z.parasiluri in this area, hence, Chongqing was a new distribution area of this species.Phylogenetically, Myxidiidae was composed of seawater branch and freshwater branch;Myxidiumand Zschokkellaclustered together with each other which showed the non-monophylogeny and close genetic relationships of two genara.
