    铁皮石斛蒴果的离体保存

    Research on Conservation of Dendrobium Candidum Capsule in Vitro

    • 摘要: 以铁皮石斛杂交蒴果为材料,研究在不同保存条件下蒴果内种子的萌发率和萌发后周长变化情况,目的是筛选出铁皮石斛蒴果离体保存的适宜条件.结果表明:4℃冰箱保存30、60、150 d的蒴果情况较好; 种子接种于1/2MS + NAA0.2 mg/L+土豆汁150 g/L培养基上萌发较好,萌发率平均为81.77%,萌发10 d的种子周长较其他处理大,可作为铁皮石斛蒴果采收后短期保存的方法.

       

      Abstract: Using the hybrid capsule of Dendrobium candidumas the material, the germination and perimeter of its seeds were studied in different preservation conditions in order to find out about the suitable conditions for vitro conservation. The result showed that the optimal conservation condition is when it is kept for 30 d, 60 d and 150 d under the temperature of 4℃ in refrigerator. The seeds is found to have an average germination rate of 81.77% when it is planted in the substrate of 1/2MS + NAA0.2 mg/L+ potato juice 150 g/L. After 10 of germination, the perimeter is larger, compared with the effect by other treating methods. It proves that such a method can be used as a method for short-term preservation of Dendrobium Candidum Capsule in Vitro after harvest.
